国内筛选的脆弱类杆菌荧光抗体的评价

摘    要:通过筛选得到5株脆弱类杆菌(Bacteroides fragilis,Bf),用作者改进后的方法免疫NZW兔,获得凝集效价为1∶1280~1∶5120的五份免疫血清,并制成荧光抗体。其中SFMC10012株的荧光抗体可使100%(69/69)的同种细菌染上4~+荧光,除1株多形类杆菌(B.thetaiotaomicron)染上荧光外,未发现与其它细菌有2~+以上的交叉荧光染色。SFMC 10012菌株的发现,可简化脆弱类杆菌多价荧光抗体的制备工艺,便于生产。

Abstract:A modified method was used to produce hyperimmune sera from NZW rabbits with 5 carefully selected strains of B. fragilis. Tube agglutination tests showed their titers to be 1280-5120 respectively. Five fluoresoein isothiooyanate conjugates were prepared. Among them, the conjugate against SFMO 10012 produced 100% (69/69) staining of homologous strains with a 4+ fluorescence. It did not cross react with bacteria other than B. fragilis excepting one strain of B. thetaiotaomicron. The discovery of this strain of B. fragilis simplified the preparation and quality control of B. fragilis polyvalent conjugates.
