虎杖苷对大鼠急性脑缺血再灌注损伤的保护作用

摘    要:
目的:观察虎杖苷注射液对大鼠急性全脑缺血再灌注损伤的保护作用。方法:采用双侧颈总动脉结扎法制备大鼠急性全脑缺血再灌注损伤模型,观察虎杖苷注射液对大鼠脑含水量,超氧化物歧化酶、单胺氧化酶的活性和丙二醛、乳酸含量的影响。结果:与模型组相比,虎杖苷注射液(7.5mg/kg,15mg/kg和30mg/kg)可显著改善脑水肿、减少过氧化脂质的形成,减少乳酸的聚积,并对单胺氧化酶有抑制作用,其作用强度与剂量有一定关系。结论:虎杖苷注射液对大鼠急性全脑缺血再灌注损伤有明显的保护作用。

Abstract:
Objective:Aim to study the cerebral pr otective effects of Polydatin (Pol) on cerebral ischemia-reperfusion injury in r ats.Methods:The rats were divided into these groups: model gro up, sham group, and 3 Pol groups (7.5 mg/kg, 15 mg/kg, and 30 mg/kg). The model of acute reperfusion injury after cerebral ischemia in rats was made by clamping bilateral carotid arteries. 1 hour after the ischemia occurred, the Pol and sal ine were administered via sublingual vena to the rats respectively and the ische mic condition lasted for another hour with a subsequent reperfusion of 24 hours. The rats were decapitated for measurement of water contents of whole hemisphere s, cerebral superoxide dismutase (SOD) activity, malonyldialdehyde (MDA) content , lactic acid (LA) content and monoamine oxidase (MAO) activity.Results:Compared with sham group and 3 Pol groups, cerebra l water content, MDA content, LA content and MAO activity were much higher in mo del group(P<0.01). The SOD activity was lower in model group with a signifi cant difference between sham and 3 Pol groups(P<0.01). Conclusion:Polydatin has a great protective effect on cere bral ischemia-reperfusion injury.
