摘    要:目的建立大鼠血浆中双嘧达莫的RP-HPLC检测方法。方法采用RP-HPLC法测定大鼠血浆中药物浓度,色谱柱为Diamonsil ODS C18(200mm×4.6mm,5μm),流动相为甲醇-pH6.5磷酸二氢钾缓冲液(60∶40,v/v)。结果在0.10~10.0μg/mL范围内,双嘧达莫与内标峰面积比值与浓度线性关系良好(r=0.997),最低检测限为0.05μg/mL,绝对回收率为84.3%~92.3%,准确度为99.9%~103.1%,日内、日间RSD均<10%。结论本测定方法灵敏、准确、简便,适合于双嘧达莫的药代动力学研究。

Abstract:Objective To develop a simple,sensitive and rapid method for the determination of dipyridamolein rats plasma.Methods The plasma concentration of dipyridamole was determined by RP-HPLC method,the analysis involved a 200 mm×4.6 mm column packed with C18 (5 μm),the mobile phase consisted of methyl alcohol-pH 6.5 potassium dihydrogen phosphate buffer (60∶40).Results A good linearity was obtained in the range of 0.10~10.0 μg/mL(r=0.997),the recovery was between 84.3%~92.3%,the accuracy was between 99.9%~103.1%,the RSD of within-day and between-day were less than 10%,the minimal detectable concentration in rats plasma was 0.05 μg/mL.Conclusion The method is sensitive,accurate and simple for the determination of dipyridamole in rats plasma,it was suitable for the pharmacokinetics study of dipyridamole.
