接骨中药含药血清对成骨细胞生物功能的影响

目的 比较伤科常用单味接骨中药含药血清对成骨细胞生物学功能的影响。方法 应用MTT法、对硝基苯磷酸盐法、茜素红染色方法和RT-PCR法，比较伤科常用单味中药当归、自然铜、骨碎补和续断对体外培养成骨细胞的增殖、碱性磷酸酶分泌、矿化结节形成和mRNA表达的影响。结果 单味中药当归对早期增殖期的成骨细胞有明显的促增殖和分化的作用；当归和自然铜能增加成骨细胞矿化期矿化结节的形成；当归和续断能上调BMP-2和IGF-ImRNA的表达。结论 单味接骨中药当归具有刺激体外培养成骨细胞增殖和分化的功能。

Effects of Chinese herb medicine on the biological functions of cultured osteoblasts in vitro

Objective To compare the effects of single Chinese herb medicine on cultured osteoblasts in vitro. Methods MTT, PNPP, ARS and RT-PCR were used to observe the proliferation, activity of ALP, the number of mineral nodes and the expression of BMP-2 and IGF- I of mRNA in osteoblasts cultured in vitro. Results It was found that Angelica sinensis has effects on stimulating, ALP activity in the early time of proliferation, Angelica sinen-sis and Pyrite can increase the number of mineral nodes in the mineralization time, Angelica sinensis and Dipsacus japouicus can promote the expression of BMP-2 and IGF- I mRNA of cultured osteoblasts in vitro. Conclusion Single Chinese herb medicine, Angelica sinensis, has effects on stimulating the proliferation, differentiation and maturation of cultured osteoblasts in vitro.
