连翘化学成分与抗氧化活性研究

摘    要:
目的:对中药连翘进行化学成分的分离与鉴定,并对连翘酯苷A的抗氧化活性进行研究.方法:连翘85%的乙醇提取物经D101大孔树脂,以水和不同体积分数乙醇洗脱,60%与30%乙醇洗脱部分采用硅胶柱色谱、凝胶色谱和制备液相等手段纯化得到6个化合物;利用理化和谱学分析(NMR,ESI-MS)的方法进行结构鉴定.利用DMPD+法和DPPH法测试连翘酯苷A的抗氧化活性.结果:6个化合物被分别鉴定为连翘酯苷A(forsythoside A)(1)、木通苯乙醇B(calceolarioside B)(2)、芦丁(rutin)(3)、连翘苷(forsythin) (4)、异橄榄脂素(isoolivil)(5)、胡萝卜苷(daucosterol)(6).结论:连翘酯苷A有很好的抗氧化活性.

Abstract:
Objective: To study the chemical constituents of Forsythia suspensa and antioxidant activity of forsythoside A. Method: The compounds were isolated and purified by column chromatography on silica gel Sephadex LH-20 and preparative HPLC, and their structures were identified by physicochemical properties and spectral analysis. DMPD·+and DPPH experiments were conducted to examine the antioxidant activity of forsythoside A. Result: Six compounds were isolated from the fruits of F. suspensa. The structures of the compounds were determined as forsythoside A (1), calceolarioside B (2), rutin (3) and forsythin (4), isoolivil (5), daucosterol (6), respectively. Conclusion: Forsythoside A showed excellent antioxidant activity.
