STUDY OF BACTERICIDAL MECHANISM OF PERACETIC ACID AGAINST SPORES OF BACILLUS SUBTILIS VAR. NIGER |

Abstract: | ![]() Spores of Ba cillus subtilis var. niger were exposed to 0.2%, 1% and 2% peracetic acid at 25℃. The structural changes induced by peracetic acid were examined with electronmicroscope. The ultrastructure of spores showed not only the progressive disintegr ation of the coat and cortex, but also disintegration and lysis of the core.Spores of B. subtilis var. niger were labeled by ~3H-TdR, ~3H-Urd or ~3H-Leu in the synthetic medium, then exposed to 0.2%, 1% or 2% peracetic acid at appropriate intervals. The radioactivity of their filtrates and the amount of dipicolinic acid (DPA) of the spores were measured. The results showed that the spore constituents, such as DNA, RNA, DPA and amino acids leaked out and the leakages were inversely proportional to the number of survived spores.The above results suggest that peracetic acid damages the permeability barrier of spore and destroys the cytoplasm of spore core. This may be the mechanism of the bactericidal effect of peracetic acid. |
