摘    要:目的 探讨不同浓度全反式维甲酸(ATRA)对食管癌细胞系EC109的生长抑制作用及其机理.方法 不同浓度ATRA作用于细胞EC109,采用MTT法检测细胞生长;通过流式细胞仪检测和DNA梯状电泳证实凋亡存在;用RT-PCR方法检测凋亡诱导过程中p53、bcl-2和bax基因表达变化.结果 ATRA呈剂量及时间依赖性抑制细胞生长;琼脂糖电泳呈现凋亡DNA梯状条带;流式细胞仪分析存在明显的凋亡峰;ATRA处理组bax和p53基因表达上调,但bcl-2基因表达下调.结论 ATRA具有抑制食管癌细胞株EC109生长作用,其机理可能是诱导细胞凋亡,其分子机制可能与bcl-2/bax和p53的表达相关.

Abstract:Objective To study the effects of a wide range of concentration of all-trans retinoic acid(ATRA)on esophageal cancer cell EC109.Methods The cell growth activity was detected by MTT assay after treatment with different concentration;the apoptosis was detected by flow cytometry assay and DNA Gel electrophoresis;the expressions of p53 and bcl-2/bax mRNA were assessed by RT-PCR.Results The cell growth was depressed by ATRA depending on concentration and duration obviously;the presented apoptosis features were observed;DNA gel electrophoresis showed DNA ladder;flow cytometry analysis showed an obvious apoptosis peak;the expressions of bax and p53 were up-regulated while the expression of bcl-2 was down-regulated in the treated group.Conclusions ATRA inhibits the growth of esophageal cancer cell EC109 obviously through inducing cell apoptosis,its molecular mechanism may be related with the expressions of bcl-2/bax and p53.
