摘    要:目的:表达严重急性呼吸综合征(SARS)冠状病毒S蛋白的抗原表位序列,为研制新型的SARS病毒疫苗提供新思路。方法:应用生物信息学分析软件分析SARS冠状病毒S蛋白的抗原表位序列,设计全新的抗原表位编码基因序列,构建表达载体并在大肠杆菌中表达该基因。结果:设计并合成了SARS冠状病毒S蛋白的抗原表位序列的新基因序列,在大肠杆菌中实现了该基因的高表达。结论:可以重新设计新基因产生新型候选重组疫苗。

Abstract:Objective:To design a new gene encoding the antigen epitope of the spike protein(S protein) of severe acute respiratory syndrome coronavirus (SARS-CoV) and express the new gene. Methods:By analyzing the antigen epitope of SARS-CoV S protein with antigenic peptides prediction software,a new gene was designed,and its expression vector was constructed. Then the gene was expressed in Escherichia coli (E coli). Results:The new gene encoding the antigen epitope of SARS-CoV S protein was designed and synthesized successfully,and it was highly expressed in E coli. Conclusion:The new gene encoding the antigen epitope of SARS-CoV S protein could be designed,which provides basis for the development of the new type of recombinant vaccine candidate.
