广州管圆线虫特异性基因的检测

摘    要:目的广州管圆线虫特异性基因的探索和检测以及其应用诊断价值。方法根据广州管圆线虫的特异性rRNA基因的序列特点,设计引物,对广州管圆线虫成虫、Ⅲ期幼虫及Ⅰ期幼虫及虫卵的DNA进行PCR扩增,并对扩增产物进行测序。结果经电泳和DNA序列测定,广州管圆线虫各期虫体及虫卵的rRNA基因片段有特异性。结论广州管圆线虫的大亚基rRNA基因是细胞中含量最多,且结构保守,编码为多拷贝的基因,易于检测发现,具有进行诊断广州管圆线虫的价值。

Abstract:In order to detect the specific subunit rRNA gene of Angiostrongylus cantonensis and evaluate the diagnosis for it the DNA of A.cantonensis from the adults and larvae of III stage were amplified with PCR,and the products were sequenced.The primers were designed according to the specific subunit rRNA gene of A.cantonensis.Results showed,with electrophoretic analysis and DNA sequence analysis,the rRNA segment was specific in different growing stages of the worm,as well as in the eggs.In conclusion,as subunit rRNA gene is the majority gene in cell with conserved structure and multiple copies of the gene encoding,it's easy to detect and its value in A.cantonensis diagnoses is significant.
