摘    要:该研究建立高效液相色谱串联四极杆飞行时间质谱法(HPLC-Q-TOF-MS /MS)以及HPLC-DAD法从定性定量2个方面分别探讨中药白芍饮片与赤芍饮片化学成分以及含量差异,定性采用Q-TOF-MS法,负离子模式扫描,鉴定出白芍38个成分,赤芍30个成分.定量采用HPLC-DAD法,分析14种主要成分(没食子酸、原儿茶酸、芍药苷亚硫酸酯、原儿茶醛、没食子酸甲酯、氧化芍药苷、儿茶素、芍药内酯苷、芍药苷、没食子酸乙酯、苯甲酸、五没食子酰葡萄糖、苯甲酰芍药苷、丹皮酚)的含量,在考察的浓度范围内测定的成分线性关系良好(r> 0.999 0),回收率(n=6)均在96.0% ~ 101.2%,RSD均小于2.78%,并且8种成分在白芍和赤芍中分布存有差异.结果表明,通过HPLC-DAD-Q-TOF-MS/MS联用技术,为鉴定白芍与赤芍的成分提供了一种快速、高效的定性、定量分析方法,为综合评价白芍与赤芍的质量提供参考.

Abstract:In order to explore the differences of chemical constituents of Paeoniae Radix Alba and Paeoniae Radix Rubra, a qualitative analytical method of liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(HPLC-Q-TOF-MS/MS) was developed for identification of multi-constituents and an HPLC-DAD analytical method was developed for simultaneously determining 14 major compounds(gallic acid, protocatechuic acid, paeoniflorin sulfonate, protocatechuic aldehyde, methyl gallate, oxypaeoniflorin, catechin, albiflorin, and paeoniflorin, ethyl gallate, benzoic acid, pentagaloylglucose, benzoyl-paeoniflorin, and paeonol) in Paeoniae Radix Alba and Paeoniae Radix Rubra. Q-TOF/MS qualitative analysis was performed under negative ion mode and inferred 38 components of Paeoniae Radix Alba and 30 components of Paeoniae Radix Rubra. HPLC-DAD quantitative method result showed the contents of 8 ingredients were different between Paeoniae Radix Alba and Paeoniae Radix Rubra. The results indicated that the new approach was applicable in qualitative and quantitative quality control of Paeoniae Radix Alba and Paeoniae Radix Rubra.
