胰岛素样因子3在邻苯二甲酸二(2-乙基)己酯诱导隐睾发生过程中的表达分析

摘    要:目的:研究环境内分泌干扰物邻苯二甲酸二(2-乙已基)酯(Di-(2-ethylhexyl)phthalate,DEHP)作用于孕鼠后,其胎鼠睾丸组织中胰岛素样因子3(Insulin like factor3,INSL3)mRNA和蛋白质的表达情况;探讨DEHP诱发小鼠隐睾的分子机制.方法:妊娠KM小鼠随机分为3组:正常对照组、玉米油对照组和DEHP实验组.玉米油对照组和DEHP实验组自妊娠12.5d到妊娠18d分别持续经口予以玉米油和DEHP500mg/(kg.d),正常对照组不予灌药.结果:正常对照组和玉米油对照组睾丸组织中INSL3 mRNA和蛋白质的表达均无差异(P〉0.05);DEHP实验组INSL3 mRNA的表达量约为对照组的1/3,与对照组相比其差异具有显著性(P〈0.05);实验组INSL3蛋白质的表达量约为对照组的1/4,与对照组相比其差异具有显著性(P〈0.05).结论:环境内分泌干扰物DEHP下调睾丸间质细胞INSL3 mRNA和蛋白质的表达,影响睾丸引带发育,可能是DEHP诱发隐睾的分子机制之一.

Abstract:AIM:To investigate the expression of Insulin like factor 3(INSL3)in fetal mice exposed to Di-(2-ethylhexyl)phthalate(DEHP)in pregnancy.METHODS:KM mice were divided into 3 groups randomly:normal control group,corn oil group and DEHP-exposed group.The last two groups were administrated respectively with corn oil and DEHP 500 mg/(kg·d)to the pregnant mice from gestation day 12.5(GD12.5)to GD18,and normal group with nothing.RESULTS:Expression of INSL3 mRNA and protein in normal and corn group were no significance(P〉0.05).Expression of INSL3 mRNA in DEHP-exposed group was about one-third of that in normal group,which was significantly decreased(P〈0.05).Similarly,expression of INSL3 protein in DEHP-exposed group was about quarter of that in normal group,which was also significantly decreased(P〈0.05).CONCLUSION:DEHP as one of environmental endocrine disruptors,can down-regulate the mRNA and protein expression of INSL3 and interfere the gubernaculum development.It may be a possible mechanism that DEHP causes cryptorchidism.
