异体和自体骨髓源神经干细胞周围神经移植实验研究

摘    要:[目的]研究异体和自体骨髓源性神经十细胞移植于周围神经后神经修复情况,以及所移植神经干细胞能否以分化神经元的形式存活。[方法]取新西兰人白兔骨髓基质细胞(BMSCS)体外培养及诱导分化为神经干细胞,采用Brdu标记细胞和免疫细胞化学SABC法检测观察培养细胞:随机选择同种异体BMSCS移植、自体BMSCS移植和未移植组。[结果]异体和处自体移植区生K的神经纤维均显波纹状排列,神经纤维密度比正常神经低但形态近似,可见有髓神经纤维的郎飞氏结,可见到神经纤维间隙的黏液基质及少量成纤维细胞,但均未见分化神经元样的细胞存在;未移植侧生K的神经纤维稀疏,可见到比较多的断裂和不连续,基质黏液变明显;异体移植组神经纤维生K的走行序列比自体移植组神经纤维紊乱,可见到反应性增生。[结论]异体和白体骨髓源性神经干细胞周围神经缺损区移植均有利于神经纤维的修复,异体比自体移植后神经纤生长较差但差别较小;本方法神经干细胞朱见以分化神经元的形式存活。

Abstract:[Objective] To investigate the repairing effect of grafted neural stem cells from allo-and self-body marrow on the peripheral nerve and discuss if they can survive in the form of differentiated neural cells.[Method]The bone marrow stroma-cells from New Zealand rabbits were cultured and induced into stem cells. BrdU and SABC methods were adopted to mark and detect them. The allo-body grafting group, self-body grafting group and control group were made respectively from random selection.[Result] Nerve fibers of both allo-and self-body grafting area showed the array of ripple, which was the same shape as the normal fibers except for lower density. Node of ranvier of myelinated nerve fibers and mucus matrix between the inter-space of nerve fibers and little fibroblast were seen. The nerve fibers of none-grafting area were rarefied, discontinuous and many disruptions were found. The walk sequence of nerve fiber of allo-grafting was more disorder than that of self-grafting. Differentiated cells were not found in both allo-and self-grafting area. [Conclusion]Both neural stem cells from allo-and self-body bone marrow are helpful to repair peripheral nerve, the living condition of allo-body stem cells is worse than that of self-dody stem cells. Neither of them survive in differentiated form.
