甘油二酯食用油对大鼠肝脏功能的影响

摘    要:
目的探讨甘油二酯食用油对大鼠肝脏功能的影响。方法将45只雄性SD大鼠随机分为正常对照组(NG)(n=10)、甘油二酯(DG)组(n=17)和甘油三酯(TGL)组(n=18)。大鼠喂养12周,每周称体重,于4、8、12周末禁食眼眶静脉采血,检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆固醇(TC)、总甘油三酯(TG)和总胆汁酸(TBA)水平;取大鼠肝脏作石蜡切片,HE染色观察各组肝脏组织变化。结果大鼠血清ALT、AST、TC、TG、TBA在实验4周时各组间差异均无显著性(P〉0.05);8周时TGL组除TG外各血脂指标均显著高于NG组(P〈0.05),除AST外各血脂指标均显著高于DG组(P〈0.05,P〈0.01);12周时TGL组各血脂指标均显著高于NG组(P〈0.05,P〈0.01),除AST、TBA外各血脂指标均显著高于DG组(P〈0.05,P〈0.01)。大鼠肝脏病理切片HE染色光镜下可见TGL组肝小叶结构破坏,细胞质内可见大小不等、数量不一的圆形脂肪空泡,细胞核被挤到一侧;DG组肝细胞浊肿明显减轻,有散在炎细胞浸润,肝细胞内偶有小脂滴。结论甘油二酯食用油不会引起大鼠肝脏功能损害。

Abstract:
Objective To investigate the effect of diacylglycerol oil on the liver functions in rats. Methods Totally 45 male SD rats with art average weight of (130 ± 10) g were randomly divided into 3 groups: normal control group ( n = 10), diacylglycerol (DG) group ( n = 17 ) and triacylglycerol (TGL) group ( n = 18 ). Rats were fed for 12 weeks and weighed every week. Blood was taken from eye sockets at the 4 th, 8 th, and 12 th week to detect alanine aminotransferase (ALT) , aspartate aminotransferase (AST) , total cholesterol (TC) , total triaeylglycerol (TG), and total bile acid (TBA). The rats' livers were harvested for paraffin section and light microscopy with HE staining. Results The serum levels of ALT, AST, TC, TG, and TBA were significantly increased in the TGL ~oup compared with the normal control group and was not significant different between the DG group and the normal control group. Light microscopy showed typical hepatic injuries of chronic fatty liver in TGL group while no obvious changes were found in DG group. Rats in the normal group showed normal histology of liver. Conclusion The intake of diacylglycerol oil does not propose damage to the liver functions in rats.
