半乳糖化重组人生长激素和重组人生长激素在小鼠体内的药物动力学

摘    要:目的:比较半乳糖化重组人生长激素和重组人生长激素在小鼠体内的药物动力学特征。方法:用^125I标记Gal-rhGH和rhGH、通过小鼠尾静脉iv,测定血和肝中相对放射性随时间的变化,以3P87药物动力学程序进行模型拟合和参数求算。结果:^125I-rhGH-Gal在血中的相对放射性呈现双峰,其体内药行动力学特征与^125I-rhCH明显不同,进一步证实了Gal-rhGH的肝靶向性。

Abstract:AIM : To compare the pharmacokinetics of galactosed recombinant human growth hormone (Gal-rhGh) and recombinant human growth hormone(rhGH). METHODS: ~(125)I-Gal-rhGH and 125I -rhGH was labeled with chloramine-T method. They were given to mice by iv, respectively. The pharmacokinetics of them were investigated by computer program 3P87. RESULTS: The relative radioactivity of 125I -Gal-rhGH had double peaks in blood. There were significant different pharmacokinetic characters between 125I -Gal-rhGH and 1251-rhGH. CONCLUSION: The pharmacokinetic difference between 125I-Gal-rhGH and 125I -rhGH is successfully compared by isotope tracer labeling method.
