成人骨髓间充质干细胞体外培养扩增的初步研究

目的:建立成人骨髓间充质干细胞(mesenchymalstemcells,MSCs)体外培养和扩增的方法,探讨其生物学特性,建立稳定的MSCs体外培养扩增体系。方法:取正常成人骨髓,用含10%新生牛血清的LG-DMEM培养液培养、扩增后,进行倒置显微镜观察,测定生长曲线,流式细胞仪行细胞表面抗原检测。结果:培养扩增获取的成人骨髓MSCs形态均一,增殖能力强。所获得细胞CD44表达阳性,CD19、CD15、CD45、CD34、CD38、CD4、CD8表达阴性。结论:所建立的分离和培养方法可获取骨髓黏附细胞中一组独特的细胞群,具有MSCs的生物学特性。
