阿尼西坦对VD大鼠学习记忆功能和海马齿状回长时程增强的影响

摘    要:
目的:研究阿尼西坦(Ani)对血管性痴呆(VD)大鼠学习记忆功能和海马齿状回长时程增强(LTP)的影响.方法:设对照组、VD模型组、Ani组,改良Pulsinelli四血管闭塞法建立VD大鼠模型,Ani组在模型制备成功后用Ani 300 mg/(kg*d)连续灌胃4 w,3组大鼠水迷宫实验检测学习记忆能力,在体海马齿状回记录LTP观察学习记忆的电生理改变.结果:与VD模型组比较,Ani组平均逃避潜伏期明显缩短(P<0.01),原站台象限活动时间明显延长(P<0.01),穿越站台次数明显增多(P<0.01);高频刺激(HFS)后PS潜伏期缩短程度明显增大(P<0.05或P<0.01),EPSP斜率、 PS幅值明显增大(P<0.05或P< 0.01).结论:阿尼西坦通过影响突触可塑性的某些环节易化在体海马齿状回LTP的诱导和维持,这可能是其改善VD大鼠学习记忆能力的作用机制之一.

Abstract:
Objective: To study the effect of aniracetam to the learning capability,memory and hippocampal dentate gyrus LTP in vascular dementia model rats in vivo.Methods: Sixty SD rats were randomly divided into three groups: control group,model group and Ani group.The experimental vascular dementia animal model was established with improved four vessel occlusion method.Then,aniracetams were instilled into rats of group Ani by intragastric administration every day for 4 weeks.The capabilities of learning and memory of the experimental animals were assessed through Morris Water Maze Behavior Test.Changes of LTP in hippocampal dentate gyrus were observed as the electrophysiological index of learning and memory.Results: Compared with the model group,the average escape latency in place navigation test were shorter(P<0.01),the swimming time in third quadrant in spatial probe test were longer(P<0.01) and times of traversing platform were increased(P<0.01) in experimental rats of group Ani;After HFS,PS latency shortened(P<0.05 or P<0.01),EPSP slope and PS amplitude were increased(P<0.05 or P<0.01).Conclusions: Aniracetam could facilitate the induction and sustainment of LTP in hippocampal dentate gyrus through influencing some taches of synaptic plasticity in vivo.
