东方田鼠血清被动转移抗日本血吸虫的保护力研究

摘    要:
目的 探讨被动转移东方田鼠血清抗日本血吸虫感染力及其作用机理。 方法 将东方田鼠血清通过尾静脉注射途径被动转移至小鼠 ,观察攻击感染日本血吸虫尾蚴后的减虫率、减卵率 ,并采用ELISA分别检测抗日本血吸虫童虫、成虫和虫卵的 8种抗体。 结果 与生理盐水对照组比较 ,东方田鼠血清受体小鼠获得的减虫率为 3 6.2 % ,减卵率为 5 4.0 % ;血清IgE、IgM、IgG及其亚类抗体均有升高 ,其中抗童虫抗原的IgG1抗体水平增幅最大。各试验组小鼠血清抗体水平差异均无显著性 (P >0 .0 5 )。 结论 东方田鼠抗日本血吸虫感染的天然抵抗力可通过血清被动转移至小鼠 ,使之获得部分抗血吸虫感染的保护力 ,该保护力可能是通过同时诱导受体小鼠Th1和Th2型免疫应答发挥作用的

Abstract:
Objective To study the protection against Schistosoma japonicum induced by passively transferred sera from Microtus fotis in the mouse. Methods Sera from M. fotis, Rat and BALB/c mice were injected to recipient mice via the tail vein, respectively. Resistance was observed by the reduction in the number of worms recovered from mice challenged with S. japonicum cercariae and the levels of eight antibodies include IgG subclass anti- soluble schistosomulum antigen (SSA), soluble adult worm antigen (SAWA) or soluble egg antigen (SEA) detected by ELISA. Results Compared with control mice injected with physiological saline solution, the recipient mice injected with sera from M. fotis exhibited reductions in adult worm burdens of 36.2% and in egg burdens of 54.0%.The antibody level of IgE, IgM, IgG and its subclass in all the recipient mice was found to be higher than that in control mice, and the level of IgG1 response to SSA to be the highest of all antibodies. But, there were no significant differences in antibody levels of the recipient mice. Conclusion The resistance of M. fotis against S. japonicum can partly protect mice by passive transfer sera, but this kind of protection probably acted through the induction of both Th1- and Th2-type immune responses in the recipient mice.
