类胰岛素一号增长因子微球对成骨细胞功能的影响

摘    要:目的:研究不同浓度类胰岛素一号增长因子(IG F‐1)微球对成骨细胞功能的影响。方法缓释微球制作采用乳化冷凝聚合交联法;根据微球的药代动力学特性,将载不同浓度的IGF‐1明胶微球分为0.25,2.5,25,250 ng/mg组及空白对照组,通过细胞增殖和分化实验评价不同浓度IGF‐1微球对成骨细胞功能的影响。结果载不同浓度IGF‐1的微球对细胞增殖的影响有显著性差异,0.25,2.5,25 ng/mg组对成骨细胞均具有显著的促增殖作用,其中25 ng/mg组的促增殖作用最为显著。载不同浓度IGF‐1的微球对细胞分化的影响有显著性差异,其中25 ng/mg组的促分化作用最为显著。结论 IGF‐1明胶微球有显著促进成骨细胞增殖和分化的作用,其最佳浓度为25 ng/mg。

Abstract:Objective To evaluate the effects of gelatin microspheres loaded with different concen‐trations of IGF‐1 on the function of osteoblast cells in vitro . Methods The improved emulsified cold condensation method was employed to fabricate the gelatin microspheres loaded with following five concen‐tration groups of IGF‐1 :0 .25 ,2 .5 ,25 ,250 ,0 (negative control) ng/mg . The effects of loaded gelatin microspheres on proliferation and differentiation of MG63 were then checked in vitro . Result Significant different effects of gelatin microspheres loaded with different concentrations of IGF‐1 on osteoblast cells’ proliferation were evident . The proliferation amount of the cells was in the following order :group 25 ng/mg>group 2 .5 ng/mg>group 0 .25 ng/mg >group 0 ng/mg>group 250 ng/mg(P<0 .05) . Sim‐ilarly ,significant different effects on osteoblast cells’ differentiation were found in the following fashion :alkaline phosphatase (ALP) in group 25 ng/mg > groups 2 .5 ,250 and 0 .25 ng/mg > group 0 ng/mg (P<0 .05) . Conclusion Gelatin microspheres loading with IGF‐1 had positive effects on the function of osteoblast cells . The preferred concentration of IGF‐1 on the gelatin microspheres is 25 ng/mg .
