青荚叶指纹图谱的研究

摘    要:目的建立青荚叶的高效液相(HPLC)指纹图谱,为其制药、采收、利用及质量评价提供依据。方法以木犀草素为对照品,采用反相高效液相色谱法测定10批不同商家的青荚叶,绘制色谱图,确立参照指纹图谱,并用中药指纹图谱相似度评价系统,计算其相似度。色谱条件:色谱柱:ODS C18(250mm×4.6mm,5μm);流动相:甲醇-2mL·L~(-1)磷酸水溶液;柱温:25℃;流速:1.0mL·min~(-1);检测波长:350nm;记录时间:30min。结果共标示出青荚叶的5个特征指纹峰;各批样品相似度介于0.80~0.99之间。表明不同批次的青荚叶组成相似,但含量稍有差异。结论所建立的青荚叶HPLC指纹图谱测定方法稳定性及重复性较好,为青荚叶的应用及质量评价提供依据。关键词:青荚叶;反相高效液相色谱法;指纹图谱

HPLC fingerprint of Helwingia j aponica
Abstract:Objective To establish HPLC fingerprint of Helwingia japonica and to provide a reference for harvest ,application and quality evaluation of Helwingia japonica .Methods RP‐HPLC method was used to determine the content of 10 batches Helwingia japonica from different habitats with rutin and quercetin as control .The similarity was calculated using fingerprint similarity eval‐uation system .The separation was performed on a Hypersil ODS C18 (250 mm × 4 .6 mm ,5 μm) column with mobile phase consis‐ted of methanol‐2 mL · L -1 phosphoric acid solution(gradient elution)at a flow rate of 1 .0 mL · min-1 lasting for 30 min .The UV detection wavelength was set at 350 nm and column temperature was at 25 ℃ .Results The results showed that 5 peaks were rec‐ognized as common peaks .The similarity index of 10 batches Helwingia japonica samples were between 0 .80~0 .99 .The results indicated the chemical components were similar but the contents of the compounds was different .Conclusion The established HPLC fingerprint of Helwingia japonica is stable ,reliable and reproducible ,which is valuable for harvest ,application and quality evaluation of Helwingia japonica .
