复发性生殖器疱疹患者外周血IL-12与Th1/Th2细胞因子的检测

目的检测复发性生殖器疱疹(RGH)患者不同病期外周血CD4+T细胞内IL-12,IFN-,γIL-4的水平,探讨IL-12,Th1与Th2亚群在疾病中的可能作用。方法应用流式细胞仪对20例发作期、15例恢复期RGH患者和15名健康人外周血CD4+T细胞IL-12,IFN-γ和IL-4进行检测。结果发作期患者外周血IFN-γ+-CD4+T细胞百分率显著低于正常对照组(P<0.05),IL-4+-CD4+T细胞百分率明显高于正常对照组(P<0.01),Th1/Th2比值显著低于正常对照组(P<0.01),同时IL-12+-CD4+T细胞百分率显著降低(P<0.01)。恢复期患者外周血IL-12+-CD4+T细胞百分率仍显著低于正常(P<0.05)。结论RGH患者存在Th1/Th2比例失衡和IL-12水平低下,而后者可能是导致Th1/Th2比例失衡和病情反复发作的重要原因。

Detection of IL-12 and Th1/Th2 Subsets in the Peripheral Blood of Patients with Recurrent Genital Herpes

Objective To detect cytokine levels of IL-12, IFN-γ and IL-4 secreted by CD4~+T lymphocytes from patients with recurrent genital herpes(RGH) at different periods and investigate their roles in the pathogenesis of RGH.Methods Flow cytometric analysis was employed to study intracellular cytokines(IL-12, IFN-γ and IL-4) in CD4~+T lymphocytes in the peripheral blood of 20 patients with RGH at acute period,15 patients with RGH at recovery period and 15 healthy volunteers.Results Compared with healthy controls,RGH patients at acute period showed a decreased percentage of IFN-γ-producing CD4~+T lymphocytes (P<0.05),an increased percentage of IL-4-producing CD_4~+T lymphocytes and a decreased ratio of Th1/Th2(P<0.01),whereas a decreased percentage of IL-12-producing CD_4~+T lymphocytes (P<0.01).RGH Patients of recovery period showed only a decreased percentage of IL-12-producing CD_4~+T lymphocytes in comparison with healthy volunteers (P<0.05).Conclusion There is a low level of IL-12-producing CD4~+T lymphocytes and Th1/Th2 imbalance in RGH patients. Low level of IL-12-producing CD~4+T lymphocytes in RGH patients may be an important cause of recurrence and Th1/Th2 imbalance.
