米诺环素对脑缺血再灌注损伤皮层TrkA/Akt通路的影响

摘    要:目的:探讨米诺环素对脑缺血再灌注(I/R)损伤的保护机制。方法:线栓法制备大鼠局灶性脑缺血模型,腹腔给予米诺环素,评价神经缺损体征,免疫组化及Western Blot检测米诺环素对皮层神经生长因子受体TrkA/Akt通路的影响。结果:米诺环素能显著改善I/R诱导的神经缺损体征,同时能诱导皮层TrkA表达上调及Akt活化(P<0.01)。结论:米诺环素可通过上调TrkA/Akt通路,减轻I/R损伤。

Effects of minocycline on TrkA/Akt pathway induced by focal cerebral ischemia following reperfusion
Abstract:[Abstract] Objective: To investigate the underlying neurobiological mechanism of the protective effect of minocycline after temporary middle cerebral artery occlusion (tMCAO) in rats. Methods: Rats underwent 90 minutes of MCAO. Minocycline or saline placebo was administered intraperitoneally starting at reperfusion. Neurofunctional tests were carried out at 24 h after tMCAO. The effects of minocycline on TrkA/Akt pathway were detected by Western blot and immuno-histochemistry. Results: The severe neuronlogic deficits induced by tMCAO were significantly released after treated with minocycline (P<0.05), and at the same time, TrkA expressionm and Akt activity were significantly increased (P<0.05). Conclusions: Minocycline can upregulate the TrkA/Akt pathway and exert its neuroprotection against tMCAO.
