河湟谷地胃癌患者TNF-α、NF-κB P65的表达及临床意义

目的 探讨河湟谷地胃癌患者TNF-α及NF-κB P65的表达及临床意义.方法 采用ELISA法检测河湟谷地健康人、胃炎患者及胃癌患者（各20例）血清中TNF-α的表达,Western blot法检测上述胃癌患者癌组织及癌旁组织NF-κB P65蛋白的表达,分析TNF-α和NF-κB P65与胃癌发生、发展的相关性.结果 TNF-α在健康入、胃炎患者及胃癌患者血清中的表达具有差异性（P ＜0.001）,并与胃癌分期及淋巴结转移具有相关性（P＜0.05）.NF-κB P65蛋白在胃癌组织和癌旁正常组织中的表达差异具有统计学意义（P＜0.001）,并与胃癌分期及淋巴结转移有相关性（P＜0.05）.胃癌患者TNF-α和NF-κBP65的表达呈正相关（r=0.745,P＜0.01）.结论 TNF-α和NF-κB P65具有作为河湟谷地胃癌患者早期诊断的分子标志物的潜力,并且二者在胃癌发生、发展及转移过程中可能起重要协同作用.

Expression and Clinical Significance of TNF-α and NF-κB P65 in Gastric Cancer of Hehuang Valley

Objective To investigate the expression and their clinical significance of TNF-αand NF-kappa B P65 in patients with gastric cancer in Hehuang Valley.Methods The expression of TNF-αin healthy people,patients with gastritis and patients with gastric cancer （20 cases,respectively） in Hehuang Valley were detected by ELISA,and the expression of NF-κB P65 were detected in 20 patients with gastric cancer by Western blot.The correlation between the expression of TNF-α,NF-κB P65 and the occurrence,development of gastric cancer were analyzed.Results There was a significant difference in the expression of TNF-αin healthy people,patients with gastritis and patients with gastric cancer （P ＜ 0.001）,which was associated with stages and lymph node metastasis of gastric cancer （P ＜ 0.05）.A statistical difference in the expression of NF-κB P65 between gastric cancer tissues and normal tissues was demonstrated obviously （P ＜ 0.001）,which was also associated with gastric cancer stages and lymph node metastasis （P ＜ 0.05）.Expression of TNF -α and NF-κB P65 were positively correlated in gastric patients（r =0.745,P ＜0.01）.Conclusion TNF-α and NF-κB P65 may be the possible molecular markers for early diagnosis of gastric cancer in Hehuang Valley.TNF-α and NF-κB P65 may play an important synergistic role in the occurrence,development and metastasis of gastric cancer.
