LC—MS法测定人血浆中罗红霉素的含量

目的建立LC-MS法测定人血浆中罗红霉素的浓度。方法色谱柱：Lichrospher C18（dp5μm，250mm×4．6mmID）流动相：甲醇-10mmol/L醋酸铵水溶液（含1％的冰醋酸）（80：20，v／v）；检测波长为210nm，柱温：30℃；流速：1．0ml／min，取上清液直接进样，进样量为20μl。结果罗红霉素线性范围为0．00989—14．81μg/ml。罗红霉素的最低检测限为0．00989μg／ml，日内、日间RSD均〈5％，相对回收率为97．6％-104．5％，提取回收率均〉91．3％。结论方法灵敏，简便，可重复的，可用于罗红霉素的血药浓度测定。

Detection of roxithromycin in human blood plasma by RP-HPLC

Objective To establish LC-MS method for determination Cefepime in plasmal. Methods Lichrospher C18 was used with the mobile phase of methanol/10 inmol/L ammonium acetate solution（containing 1% acetic acid） （ 80/20 ,v/v） at the detection wavelength of 210 nm. The flow rate was 1.0 ml/min. Results Under the condition of this Roxithromycin in plasma was well separated, The calibration curve was linear in the range of 0. 009 89 - 14. 81 μg/ml. The limit of quantification was 0.02 μg/ml. The RSD of intra-and inter day were all less than 5%, the relative recovery was 97. 6% - 104. 5% and the extraction recovery was above 91.3%. Conclusion This validated method is sensitive,simple and repeatable enough to be used in pharmacokinetic studies.
