大鼠静脉注射乳化异氟烷的急性毒性

目的 研究大鼠尾静脉注射乳化异氟烷的急性毒性并确定其半数致死剂量(LD_(50)).方法 清洁级SD大鼠60只,按雌雄分层随机分为6组,溶媒对照组(C组)和不同剂量乳化异氟烷组(I_(1～5)组),每组10只,雌雄各半.I_(1～5)组分别单次尾静脉注射乳化异氟烷225.6、192.0、163.2、139.2、118.8 mg/ks,以心电图成一直线判断死亡,采用Bliss法计算乳化异氟烷的LD_(50)及其95%可信区间.C组单次尾静脉注射等容积30%脂肪乳.注射乳化异氟烷后观察2周.结果 大鼠单次尾静脉注射乳化异氟烷后出现翻正反射消失、呼吸抑制、心律失常或死亡;乳化异氟烷的LD_(50)为170.76 mg/kg,95%可信区间为153.00～190.56 mg/kg.结论 大鼠单次尾静脉注射乳化异氟烷的LD_(50)为170.76 mg/kg,急性毒性主要表现为呼吸循环抑制.

Acute toxicity of intravenous isoflurane in rats

Objective To investigate the acute toxicity of intravenous isoflurane and determine the median lethal dose(LD_(50))in rats.Methods Sixty SD rats were randomly divided into control group(n = 10)and different doses of isoflurane group(I_(1-5) group,n = 10,5 male,5 female).The I_(1-5) group received a bolus of intravenous isoflurane 225.6,192.0,163.2,139.2 and 118.8 mg/kg injected via tail vein over 10 seconds respectively and LD_(50) was calculated by Bliss.In control group 30% lipid emulsion was administered instead of isoflurane.The survived rats were observed for two weeks.Results The animals exhibited loss of righting reflex,cardio-respiratory depression or death.The LD_(50) of intravenous isoflurane was 170.76 mg/kg and 95% confidence interval was 153.00-190.56 mg/kg.Conclusion The LD_(50) of intravenous isoflurane was 170.76 mg/kg,and the acute toxicity of intravenous isoflurane is cardio-respiratory depression.
