动态增强磁共振评价重组人血管内皮抑素抑制人肺癌裸鼠皮下移植瘤的血管生成

摘    要:目的 探讨动态增强磁共振(DCE-MRI)评价重组人血管内皮抑素(恩度)抑制人NCI-H460肺癌裸鼠皮下移植瘤血管生成的可行性。方法 10只人NCI-H460肺癌荷瘤裸鼠,随机分为对照组和实验组,每组各5只。肿瘤接种第10天开始给药,实验组每日腹腔注射恩度0.2ml,对照组每日腹腔注射生理盐水0.2ml。连续给药7天后行DCE-MRI扫描,扫描结束后剖取肿瘤组织行CD34免疫组化染色。结果 实验组肿瘤体积为(0.249±0.138)cm,小于对照组的(0.448±0.096)cm,差异有统计学意义(=0.032)。实验组Ktrans、Kep、Ve及iAUC均值分别为(0.052±0.015)/min、(0.334±0.111)/min、0.280±0.191和11.912±7.738,均小于对照组(0.149±0.026)/min、(0.478±0.294)/min、0.558±0.052和36.463±12.534,其中Ktrans、Ve和iAUC两组差异有统计学意义(P<005)。实验组微血管密度(MVD)计数为(11.36±4.57)条/高倍视野,明显低于对照组的(20.44±3.46)条/高倍视野(<0.05)。结论 抗肿瘤血管生成药物恩度对裸鼠肺癌皮下移植瘤具有良好的抑瘤作用,DCE-MRI是评价肿瘤血管生成及抗血管生成药物疗效的一种有效手段。

Abstract:Objective To investigate the feasibility of antiangiogenic effect in lung tumor xenografts following endostar therapy by DCE-MRI.Methods Two groups of nude mice bearing human lung carcinoma were injected with phosphate-buffered saline and endostar,respectively.DCE-MRI was performed on day 7 after the treatment with endostar and thereafter tumor tissues were collected for immunohistochemisty analysis.Results Tumor volumes in experimental group were smaller than that in control group(P=0.032)[(0.249±0.138)cm3 vs.(0.448±0.096)cm3].The mean Ktrans,Kep,Ve and iAUC values of experimental group were(0.052±0.015)min-1,(0.334±0.111)min-1,(0.280±0.191)and(11.912±7.738),respectively.The mean values of control group were(0.149±0.026)min-1,(0.478±0.294)min-1,(0.558±0.052) and(36.463±12.534).The mean Ktrans,Ve and iAUC of experimental group were lower than those of control group and there were significant differences(P0.05).Microvessel density(MVD) of experimental group and control group were(11.36±4.57)item/hpf and(20.44±3.46)item/hpf,respectively.The MVD was decreased in experimental group compared with control group(P0.05).Conclusion Endostar can significantly inhibit the development of the human lung tumor xenografts.DCE-MRI is an effective method in evaluating tumor angiogenesis and the effect of antiangiogenic drugs.
