摘    要:
目的探讨miR-200c在复发性流产(recurrentmiscarriage,RM)患者绒毛组织中表达以及其对滋养细胞凋亡、侵袭及迁移能力的影响。方法使用qRT-PCR法检测RM患者及对照组(人工流产患者)的绒毛组织中miR-200c的表达情况;并以miR-200c模拟物和抑制物转染滋养细胞系HTR-8和JAR,观察其对滋养细胞凋亡、迁移及侵袭能力的影响。结果RM患者绒毛组织中miR-200c的表达水平显著高于对照组。体外予以miR-200c模拟物转染HTR-8/SVneo后,凋亡细胞比例增加,且细胞的侵袭、迁移能力显著降低;予以miRNA-200c抑制物转染后,凋亡细胞显著降低,且细胞的侵袭及迁移能力显著增加。结论miR-200c在RM患者绒毛组织中表达显著增加,其可能通过影响滋养细胞的凋亡、迁移及侵袭,从而影响胎盘发育,参与RM的发病过程。

Abstract:
Objective To investigate the expression of miR-200c in the villus of women suffering from recurrent miscarriage and its effect on apoptosis, migration and invasion of topho-blasts. Methods Expression of miR-200c in the villus of recuixent miscarriage women and control patients were detected by qRT-PCR.,HTR-8 and JAR cells were transfected with miR-200c mimic or inhibitor, and the effects of miR-200c on cell apoptosis was flow cytometrically detected, and their effect on migration and invasion were analyzed by Transwell assays. Results The expression of miR-200c in the villus of recurrent miscarriage was significantly higher than that in control group. MiR-200c upregulation promoted cell apoptosis. Conversely, the downregulation of miR-200c inhibited cell apoptosis. MiR-200c mimic inhibited the migration and invasion of trophoblasts, while miR-200c inhibitor had the opposite effects. Conclusion miR-200c could regulate apoptosis, migration and invasion of trophoblasts and might be involved in the pathogenesis of RM.
