摘    要:
目的:研究重组人促红细胞生成素对Ⅲ°烫伤大鼠心肌凋亡的保护作用。方法:采用Ⅲ°烫伤模型,大鼠随机分成烫伤组、对照组、EPO治疗组。大鼠烫伤后按每kg体重1%烫伤面积4mL等渗盐水腹腔注射,并于伤后18h处死动物。HE染色及S-P免疫组织化学染色观察心肌细胞组织结构改变和bcl-2基因表达情况。结果:EPO治疗组大鼠心肌凋亡指数显著低于烫伤组(P<0.01),bcl-2蛋白表达也显著高于烫伤组(P<0.01)。结论:EPO对烫伤后大鼠心肌细胞凋亡有较好的抑制作用,其机制可能是EPO调控了凋亡相关基因bcl-2的表达。

Abstract:
Objective.The myocardial apoptosis protective effect study of rHuEPO on burned Ⅲ degree of rat. Methods: Ⅲ degree burn model used, rats were randomly divided into burn group, the control group and treated group with EPO, Rats were killed after 18 h injury. The organizational structure changing of myocardial cells were observed by HE staining, The expression of bcl-2 gene in myocardial cells were observed by S-P immunohistochemical staining. Results: Myocardial apoptosis index in burn group with EPO treated was significantly lower than that of burn group, and the bcl-2 protein expression was significantly higher than that of burn group(P〈0.01). Conclusions. EPO on cardiac myocyte apoptosis after burn injury has a better inhibitory effect,its mechanism may be the EPO gene regulation of apoptosis-related bcl-2 expression.
