激素对菘蓝器官分化及愈伤组织诱导的影响

摘    要:目的研究激素对菘蓝器官分化及愈伤组织形成能力的影响。方法以菘蓝无菌苗的子叶和胚轴为外植体,采用含有不同植物激素配比的培养基培养菘蓝不同外植体,分析在不同激素配比的条件下诱导形成愈伤组织和分化不定根、不定芽的能力。结果在NAA 0.1 mg/L+6-BA 0.1 mg/L(14d)、NAA 0.5mg/L+6-BA 1.0 mg/L(14d)、NAA 0.5 mg/L+6-BA 0.1 mg/L(28d)三个条件下子叶的愈伤组织诱导率可达到100%;NAA(0 mg/L~0.1 mg/L)+6-BA1.0 mg/L的激素组合不定芽诱导率可达75%以上;生根培养以1/2MS培养基附加NAA 0.1 mg/L可使不定芽生根率可达100%。结论合适的激素组合,能促进菘蓝愈伤组织的形成和器官的分化。。

Abstract:Objective Study the effects of hormones on the ability of the organ differentiation and callus formation of Isatis indigotica Fort.Method With the Isatis indigotica Fort.explants of aseptic seedling cotyledon and hypocotyl,using medium containing different plant hormones on Isatis indigotica Fort.to culture different explants,analysis the ability of inducing the formation of callus and differentiation of adventitious roots and adventitious buds under different conditions of hormone combinations.Results The cotyledons callus induction rate can reach 100% under the three condition of NAA 0.1mg / L +6-BA 0.1 mg / L(14 d),NAA 0.5 mg / L +6BA 1.0 mg / L(14 d),NAA 0.5 mg / L +6-BA 0.1 mg / L(28 d);with the hormone combination of NAA(0 mg / L to 0.1 mg / L) +6-BA1.0 mg / L,adventitious bud induction rate can up to 75% above,rooting culture of 1/2MS medium supplemented with NAA 0.1 mg / L,the rooting rate can up to 100%.Conclusion The right combination of hormones can promote the callus formation and organ differentiation of Isatis indigotica Fort.
