摘    要:
目的探讨不同抗原修复方法对免疫组化染色结果的影响。方法将10种不同的单克隆抗体在7组不同抗原修复条件下,在同一实验条件、同一时间内运用EliVision免疫组化法进行实验。结果ER、PR、CD3、Bcl-2用pH9.0Tris-EDTA热修复表达效果好;C-erbB-2、PSA、CD117用10mmol/LpH6.0柠檬酸盐缓冲液热修复效果好;CK用0.125%胰蛋白酶修复表达效果好;CD34修复后表达过强;ER、PR、CD3、Bcl-2、C-erbB-2、PSA、CD117用水浴热修复的表达效果均优于微波热修复法。结论免疫组化的染色质量受不同抗原修复方法的影响。掌握抗体最佳的修复方法能极大提高免疫组化结果的阳性率及准确率。

Abstract:
Objective To study the effect of varics antigen repairing methods on the results of immunohistochemistry staining.(Methods)With Elivision immunohistochemistry method,experiments were conducted on ten deferent kinds of monoclonal antibody under the condition of seven deferent antigen repairing,and same experimental environment and time.Results Satisfied expression effects were shown when ER,PR,CD3,Bc1-2 in pH9.0Tris-EDTA heat repairing,C-erbB-2,PSA,CD117 in citrate buffer heat repairing,and CK in 0.125% of trypsin repairing solution.After being repaired,CD34 was over expressed.The expression effects of ER,PR,CD3,Bc1-2,C-erbB-2,PSA,CD117 in water heat repairing method were superior to those in microwave heat repairing method.Conclusion The quality of immunohistochemistry staining is affected by deferent antigen repairing method as well as the pH of repairing solutions.It can significantly improve the positive rate and accuracy of immunohistochemistry staining results to master the best antibody repairing method.
