结核分枝杆菌稳定L型感染检测方法的研究

摘    要:
目的探讨结核分枝杆菌稳定L型感染标本的病原学的检测方法。方法以非高渗分离培养法诱导并获得结核分枝杆菌稳定L型纯培养物,将其感染豚鼠后进行病原体分离培养.采用PCR方法检测稳定L型培养物的IS986序列并鉴定其性质。结果结核分枝杆菌稳定L型在常规细菌学方法检查中不能发现或鉴定,但采用非高渗分离培养法能够检出,5例动物病变组织有4例检测出稳定L型,PCR直接扩增动物病变组织有3例呈阳性反应。结论采用以非高渗分离培养和基因检测为主要内容的非高渗分离培养法.能够快速的检测出样品中潜伏存在的结核分枝杆菌稳定L型.可提高病原学诊断的阳性率和特异性.

Abstract:
Objective To evaluate the detection method for the stable L-form M.tuberculosis from the infected samples. Methods Pure culture of stable L-form M. tuberculosis was obtained using the non-high osmotic medium, Guinea pigs were infected with the stable L-form M. tuberculosis and the bacteria were then isolated from the culture medium. IS986 from the stable L-from bacteria was detected and analyzed by PCR. Results Although stable L-form M. tuberculosis was not detected by routine bacteriologic method, the bacteria could be isolated using the non-high osmotic medium. 4 samples could be detected the L-form of M. tuberculosis by the technique of non-high osmotic culture and 3 samples PCR results of the IS986 were positive in the 5 guinea pigs. Conclusion Method of cultivation using non-high osmotic medium and PCR are, suitable for the detection of the stable L-form of M, tuberculosis in tissue samples. These methods are specific and can improve the detection of this pathogen.
