摘    要:目的探讨电针(Ea)协同骨髓间充质干细胞(BMSCs)移植治疗对脑出血大鼠微管相关蛋白(MAP-2)表达的影响。方法采用大鼠脑尾核壳注射胶原酶与肝素制备脑出血动物模型,并分为Ea组、Ea—BMSCs组、BMSCs组和生理盐水(NS)组.分别给予Ea刺激、Ea刺激协同BMSCs移植、BMSCs移植和注射生理盐水,免疫组织化学方法观察各组在不同时间点MAP-2的表达。结果MAP-2表达在NS组各时间点之间无明显差异,在Ea组、BMSCs组和Ea—BMSCs组于第5天时达高峰。各组同-时间点比较,Ea组(1d除外)、BMSCs组和Ea-BMSCs组的MAP-2表达高于NS组(P〈0.05),Ea—BMSCs组高于Ea组及BMSCs组(P〈0.05)。结论Ea协同BMSCs移植较单纯Ea刺激或BMSCs移植更能促进脑出血大鼠MAP-2蛋白表达。提示Ea协同BMSCs移植有助于脑组织结构重塑。

Abstract:Objective To investigate the influence of electroacupncture (Ea) combined with bone marrow mesenchymal stem cells (BMSCs) transplantation on expression of mierotubule-associated protein2 (MAP-2) in rats with intracerebral hemor- rhage. Methods Intracerebral hemorrhage rat models were established by injecting collagenase and heparin into caudate nucle- us, and divided into Ea group, Ea-BMSCs group, BMSCs group and normal saline (NS) group. These groups were treated with Ea, Ea combined with BMSCs transplantation,BMSCs transplantation and injection of NS respectively. Immunohistochemical method was used to observe the expression of MAP-2 in each group at different time points. Results There was no significant differences among different time points in NS group ,but the expression of MAP-2 reached the peak on fifth day in Ea group, BM SCs group and Ea-BMSCs group. The expression of MAP-2 in Ea group (except on 1 st d), BMSCs group andEa-BMSCs group were higher than that in NS group at the same time point (/~〈0.05), and the expression of MAP-2 in Ea-BMSCs group was higher than that in Ea group and BMSCs group (P〈0.05). Conclusion Ea combined with BMSCs transplantation can promote the expression of MAP-2 protein better in rats with intracerebral hemorrhage compared with simple Ea or BMSCs transplantation, which suggests that Ea combined with BMSCs transolantation is heloful to restore brain tissue structure.
