基于标准汤剂的姜厚朴饮片UPLC特征图谱建立及生姜成分的鉴别

摘    要:目的:建立基于标准汤剂的姜厚朴饮片超高效液相色谱法(UPLC)特征图谱方法,研究不同基原和药用部位特征图谱的差异性,建立厚朴生品与炮制品的区分方法。方法:采用UPLC建立姜厚朴标准汤剂和饮片UPLC特征图谱,对姜厚朴不同基原和药用部位特征图谱的差异性进行分析,并采用液相色谱-质谱联用法(LC-MS)分析姜厚朴中生姜类成分。结果:20批姜厚朴标准汤剂、饮片有6个共有特征峰,通过对照品指认了2个特征峰,分别为厚朴酚、和厚朴酚;厚朴酚与和厚朴酚色谱峰的峰高比可能是厚朴与凹叶厚朴2种基原的区别点;姜厚朴根皮、干皮和枝皮特征图谱特征性成分含量差异有统计学意义,根皮中厚朴酚、和厚朴酚总量最高,干皮中特征峰1~4含量最高。采用LC-MS检测出姜厚朴中6-姜辣素和6-姜烯酚,并利用m/z为295和277的离子峰,成功区分厚朴与姜厚朴。结论:所建立的UPLC特征图谱方法能很好地用于姜厚朴不同基原和药用部位的差异性研究;所采用的LC-MS能有效地鉴别厚朴与姜厚朴。

Abstract:Objective:To establish specific chromatograms of ginger juice processed Magnoliae Officinalis Cortex(GJPMOC)based on standard decoction,study the differences of specific chromatograms from different origins and medicinal parts,set up the method for distinguishing the raw and processed products of Magnoliae Officinalis Cortex.Methods:The specific chromatograms of GJPMOC standard decoction and decoction pieces were established by UPLC.The differences of specific chromatograms between different origins and medicinal parts were analyzed by established specific chromatograms.The ginger constituents in GJPMOC were distinguished by LC-MS.Results:There were six common characteristic peaks in 20 batches of GJPMOC decoction pieces and standard decoction.By comparing with the reference substances,two peaks were identified,namely magnolol and honokiol.The peak height ratio of magnolol and honokiol may be the difference between two origins.The contents of characteristic components in root bark,dry bark and branch bark were significantly different.Magnolol and honokiol in root bark were the highest,and characteristic peaks 1-4 in dry bark were the highest.LC-MS was used to detect 6-gingerol and 6-shogaol in GJPMOC.The raw and processed Magnoliae Officinalis Cortex were successfully distinguished by the ionic peaks of m/z 295 and 277.Conclusion:The UPLC specific chromatograms can be used to study the difference between origins and medicinal parts.Magnoliae Officinalis Cortex and GJPMOC can be identified by LC-MS effectively.
