益母草碱对阿霉素致斑马鱼心脏毒性保护作用的初步研究

摘    要:目的观察不同浓度的盐酸益母草碱对阿霉素所致斑马鱼胚胎心脏毒性的保护作用。方法应用发育正常受精后48h的斑马鱼胚胎作为实验模型,用盐酸益母草碱+阿霉素(浓度为0+30mg/L,30mg/L+30mg/L,60mg/L+30mg/L,120mg/L+30mg/L)处理上述胚胎,分别于受精后60h、72h和96h时观察胚胎心脏的形态及心率的变化。结果各给药组以及对照组在给药后均出现心率减慢、心脏区域出血的胚胎心脏中毒现象。但益母草碱联合用药组的心脏毒性作用较小,且随着盐酸益母草碱浓度的升高,阿霉素的心脏毒性作用明显减小。结论阿霉素对斑马鱼胚胎具有心脏毒性作用.而盐酸益母草碱对阿霉素所致心脏毒性具有保护作用。

Abstract:Objective To study the protective effects of different Leonurine hydrochioride concentrations on the Adriamycin-induced cardiac toxicity of zebrafish embryos. Methods Zebrafish embryos of 48 hours post fertilization were treated with Leonurine hydrochloride (LH) and Adriamycin (concentrations:Leonurine hydrochloride + Adriamycin= (0+30) mg/L,(30+30) mg/L,(60+30) mg/L,(120+30) mg/L). The cardiac phenotypes and the heart rates at 60 h, 72 h, 96 h were assessed under the microscope. Results The embryos treated with Adriamycin showed cardiac toxicity such as the reduced heart rates and heart hemorrhage. But the toxicities were reduced in the groups combined with Leonurine hydrochloride, and the higher concentration of Leonurine hydrochloride, the greater reduction of the toxicity. Conclusion Adriamycin was toxic on the cardiac development of zebrafish embryos, and LH can protect the zebrafish embryos from the Adriamycin-induced cardiac toxicity.
