大鼠局灶性脑缺血再灌注后神经元中细丝蛋白表达的实验研究

摘    要:目的观察大脑中动脉缺血再灌注(MCAO/R)大鼠模型神经元中细丝蛋白(filamin,FLN)表达的变化情况,结合其超微结构改变,了解脑血管病变后神经元结构功能变化与FLN表达的相关性.方法制备MCAO/R大鼠模型,依照再灌注时间的不同分为2h、6h、12h、24h、3d及7d共6组,正常大鼠做对照组.按时相点取材,草酸-焦锑酸钾电镜观察,应用免疫组织化学方法染色并进行图像分析.结果除正常对照组外,其他各组神经元中均有阳性反应,12h、24h、3d组神经元中FLN表达较多,以3d时最明显.阳性神经元细胞集中在缺血半球皮质区.神经元中FLN阳性部位为胞浆的核周边部,表达均匀一致.胶质细胞中未见表达.结论再灌注损伤可引起神经元中细丝蛋白免疫阳性的表达,并与损伤时间的变化及神经元形态学改变有一定相关性,提示此现象可能与神经元的再生、修复及移行有关,有进一步研究的价值.

Abstract:Objective To observe the expression of filamin(FLN) in the neuron cells after cerebral ischemia/reperfusion(I/R) to find out the correlation between FLN and the neuron function. Methods MCAO/R model was established after rats were divided into sham operation and I/R groups. The I/R groups were divided into six subgroups according to different time from 2h to 7d. Immunohischemical staining and color image analysis were used to observe the expression. Pathological changes were observed with transmission electronic microscope. Results All groups, except the control group, express FLN in neuron in 12h, 24h and the 3rd day. Most of positive neurons could be seen in the cortex of ischemia cerebral hemisphere. No positive could be seen in glial cells. Nucleus shrinkage, mitochondrion swelling and myelin sheath lysis could be observed in transmission electronic microscope. Conclusions Pathological damage can stimulate neuron to produce FLN-like protein. The expression of this protein is related to time and neuron morphological changes. It may play a role in the regeneration and movement of neurons.
