Abstract:Objective To explore the relationship between the genetic variations of the promoter region of the MSH2 gene and the susceptibility and prognosis of lung cancer.Methods A total of 700 non-small cell lung cancer patients and 700 normal volunteers were selected for group designed case-control study.Follow-up and general data and survival data of 2 groups were collected.Polymerase chain reaction-restriction fragment length polymorphism and Taqman probe methods were used to detect genotypes of rs6757310 and rs1863332,respectively.Chi-square test was used to compare the distribution of categorical variables,and unconditional Logistic analysis was used to analyze the relationship between each genotype and genetic susceptibility to lung cancer.The Kaplan-Meier survival curve,Log-Rank test,and Cox multivariate regression model were used to evaluate the prognostic differences among patients with different genotypes.Results A case-control study showed that rs6757310 and rs1863332 variations did not change lung cancer susceptibility in general population.Further stratified analysis showed that MSH2 rs6757310 CC genotype was associated with lung cancer susceptibility in smokers(total smokers:P<0.05;cumulative smoking volume>30 packs/year:P<0.05)and led to increased susceptibility to squamous cell carcinoma(P<0.05).MSH21863332 GG only led higher susceptibility to lung cancer in elderly population(P<0.05).Survival analysis showed that the median survival time(MST)of patients with MSH2 rs6757310 CC genotype was 31 months,which was less than that of patients with CG genotype(MST:61 months)and GG genotype(MST:45 months;P<0.05).The MST of patients with MSH2 rs1863332 GG genotype was 19 months,which was less than that of patients with GT genotype(MST:39 months)and TT genotype(MST:48 months;P<0.05).Conclusion MSH2 rs6757310 and rs1863332 genetic variations may affect the susceptibility and prognosis of lung cancer.
