多指标正交试验法优选降脂复肝胶囊提取工艺

目的:优选降脂复肝胶囊的最佳提取工艺。方法:以丹参酮ⅡA和醇提浸膏率为指标,以乙醇用量、乙醇体积分数、提取时间、提取次数为考察因素,采用L_9(3~4)正交试验优化降脂复肝胶囊中丹参等3味饮片的醇提取工艺;以绿原酸、芍药苷、橙皮苷和水提浸膏率为指标,以加水量、提取时间和提取次数为考察因素,采用L_9(3~4)正交试验优化降脂复肝胶囊中赤芍等6味饮片的水提取工艺。结果:最佳醇提工艺为加6倍量的80%乙醇,提取2次,1.0 h/次;最佳水提工艺为加10倍量的水,提取2次,1.0 h/次。结论:优选的提取工艺参数合理,稳定可行,为降脂复肝胶囊提取工艺的参数确定提供了实验依据。

Optimization of Extraction Process of JiangZhi FuGan Capsules by Multi-index Orthogonal Test

Objective:To optimize the extraction technology ofJiangZhi FuGan capsules.Methods:By taking the contents of Tanshinone Ⅱ A and the yield of alcohol extract as the indexes,ethanol dosage,ethanol volume fraction,extracting time and times as the factors,alcohol extraction processes of DanShen (the root of red-rooted salvia) and other two decoction pieces in JiangZhi FuGan capsules were extracted by L9(34) orthogonal test;by taking the contents of chlorogenic acid,paeoniflorin and hesperidin and water extraction rate as the indexes,the adding water amout,extraction duration and extraction times as the factors,water extraction processes of ChiShao (radix paeoniae rubrathe) abd other five decoction pieces in JiangZhi FuGan capsules were extracted by L9 (34) orthogonal test.Results:The best alcohol extraction technology was adding six times of 80％ ethanol,extraction for twice,1.0 hour each time;the optimal water extraction technology was adding ten times of water,extraction for twice,1.0 hour each time.Conclusion:The optimized extraction process is rational,stable and reliable.It could provide experimental basis for confirming parameter of the extraction technology ofJiangZhi FuGan capsules.
