剪刀股中多糖的提取及其对糖尿病小鼠的降糖作用研究

摘    要:目的:建立剪刀股多糖的提取与含量的测定方法,探讨剪刀股多糖对四氧嘧啶诱导的糖尿病小鼠的降血糖作用。方法:通过正交试验优化剪刀股多糖提取条件,采用苯酚-硫酸法对剪刀股多糖的含量进行测定;尾静脉注射四氧嘧啶,建立糖尿病小鼠模型,并随机分为正常对照组、模型对照组及剪刀股多糖、高、中低剂量组,高、中、低剂量组分别以9、6、3 mg/10g(体质量)每天的剂量灌胃,正常对照组和模型对照组灌胃0.3 m L生理盐水,连续灌胃27天,分别于第0、3、9、15、21、27天测小鼠的血糖和体质量。结果:剪刀股多糖最佳提取条件为提取温度80℃,液料比20∶1,提取150(50×3)mi n。剪刀股多糖的平均提取率为9.64%,RSD为0.87%。给药27天,剪刀股多糖高、中、低剂量组小鼠血糖显著降低,体质量显著升高。结论:剪刀股中多糖含量较高,剪刀股多糖能较明显降低小鼠血糖值,改善糖尿病小鼠的消瘦症状。

Abstract:Objective: To establish extraction and content determination methods of polysaccharides in JianDaoGu (Ixeris debills A.Gray), and explore the glucose-lowering effects of polysaccharides extracted from JianDaoGu on mesoxalyl usea-induced diabetic mice. Methods: The extraction process of polysaccharides from JianDaoGu was optimized by orthogonal method, and the contents of polysaccharides from JianDaoGu were measured by using Phenol-sulfurie acid method. Diabetic mice models were established by tail vein injection of mesoxalyl usea, and they were randomized into normal control group, model control group, and high, moderate and low dose groups of polysaccharides extracted from JianDaoGu, the mice in high, moderate and low dose groups of polysaccharides extracted from JianDaoGu accepted intragastric administration of medicine in the dose of 9, 6 and 3 mg/10 g (body weight)each day, normal control group and model control group accepted lavage of physiological saline, 0.3 mL, for 27 days consecutively, blood glucose and body weight of the mice were detected at the zero, third, nineth,15 th, 21 st and 27 th day respectively. Results:The best conditions for polysaccharides extracted from JianDaoGu was extraction temperature 80℃, liquid material ratio was 20:1, extracting for 150 (50×3)min. Average extraction rate of polysaccharides from JianDaoGu was 9.64%, RSD was 0.87%. After medicating for 27 days, blood glucose of the mice in high, moderate and low dose groups of polysaccharides decreased notably, and body weight rasied notably. Conclusion:The contents of polysaccharides are high in JianDaoGu. Polysaccharides could nobtaly lower blood glucose of the mice and improve maransis of diabetic mice.
