参麦注射液对神经细胞凋亡及胞浆钙变化的影响

摘    要:目的:研究神经细胞缺氧/缺糖培养诱导的凋亡发生率、胞内游离钙离子变化及参麦注射液的治疗作用。方法:碘化丙啶染色,流式细胞仪定量分析细胞凋亡百分率;Fluo-3负载,流式细胞仪观察胞内钙平均荧光值变化。结果:缺氧/缺糖培养可诱导神经细胞凋亡和引起细胞内游钙离子增高,参麦注射液能抑制细胞凋亡、降低细胞内游离钙钙离子浓度。结论:凋亡是神经细胞死亡方式之一,参麦注射液抑制缺氧/缺 导的神经细胞凋亡及钙超载,有社会细胞保护作用。

Abstract:Objective: To study the neurocyte apoptosis and change of cytosolic free calcium induced by hypoxia/hypoglycemia culture in SH-SY5Y human neuroblastoma cells, and the effect of Shenmai Injection (SMI) on them. Methods: The neurocyte apoptosis rate was analysed quantitatively by flow cytometry and cytosolic calcium was determined by measuring mean fluorescent value with Fluo-3 flurometry. Results: Hypoxia/hypoglycemia could induce neurocyte apoptosis and increase the cytosolic calcium in SH-SY5Y cells. SMI could inhibit the SH-SY5Y cell apoptosis and lower the concentration of cytosolic free calcium. Conclusion: Apoptosis is one of the death pattern of nerve cells. SMI could obviously reduce the hypoxia/hypoglycemia induced apoptosis and calcium overload, thus protect the neurocytes.
