卡维地洛对大鼠缺血再灌注心肌间隙连接结构影响的研究

摘    要:目的:研究卡维地洛(CVD)对大鼠缺血再灌注损伤心肌间隙连接(GJ)结构的保护作用.方法:将24只大鼠随机分为假手术(SHAM)组、缺血再灌注(IRI)组和CVD组.CVD组以卡维地洛2mg/kg/d灌胃给药7d,SHAM组和IRI组以等量生理盐水灌胃7d.IRI组和CVD组结扎左冠状动脉前降支致大鼠心肌缺血30min后复灌4h,制作成心肌缺血再灌注损伤模型.SHAM组只穿线不结扎.再灌4h末用免疫荧光和共聚焦显微镜扫描技术观察3组心肌GJ蛋白43(CX43)的分布及组成变化.结果:与SHAM组相比,IRI组CX43-GJ结构明显异常;与IRI组比较,CVD组CX43-GJ结构基本正常.结论:卡维地洛具有保护心肌间隙连接结构的作用.

Abstract:Objective:To examine the effects of carvedilol(CVD)on the structure of myocardial gap junction(GJ) after myocardial ischmia and reperfusion injury in rats.Methods:Twenty four rats were divided randomly into three groups,sham operation(SHAM) group,myocardial ischmia and reperfusion(IRI) group and CVD group.Carvedilol(2mg/kg/d)was given intragastrically in CVD group,and equal normal saline was given in SHAM group and IRI group for seven days.Then ligated the left anterior descending branch for 30 minutes and reperfused for 4 hours to build IRI injury model in IRI and CVD group,braided the left anterior decending branch in SHAM group.After 4 hours reperfusion,the distribution and quantification of gap junctional connexin 43(CX43) was measured by immunofluorescence and laser scanning confocal microscopy.Results:Compared with SHAM group,the IRI group resulted in abnormal distribution and composition of CX43-GJ.Compared with IRI group,the CX43-GJ distribution and composition were nearly normal in the CVD group.Conclusion:Carvedilol protect the structure of CX43-GJ after myocardial ischemia-reperfusion injury.
