摘    要:目的:寻找一种适合膜片钳研究的海马神经元急性分离法。方法:采用出生7~10d的大鼠,用酶机械联合分离法制备海马神经元细胞,用全细胞膜片钳技术对其电生理学特性进行测定。结果:细胞不仅形态学完整,而且具有正常的电生理学特性,能记录到电压依赖性及配体门控GABA电流,还能记录到动作电位。结论:酶机械联合分离法简单快捷,所需酶量少,不需配置混合气,成本低,适合膜片钳的研究。

Acute Isolation of Rat Hippocampal Neurons Suitable for Patch-clamping Study

Abstract:Objective:To find out a single hippocampal neuron suitable for patch-clamp technique.Methods: Acute isolation of hippocampal neurons in 7-10-d-old neonate rats was obtained by enzyme digestion and mechanical separation technique.Electrophysiologic characteristics of hippocampal cells were studied with whole-cell patch-clamp technique.Results: Hippocampal neurons by acute isolation with good characteristics in both morphology and physiology could be obtained.Conclusion: This acute isolation of hippocampal neurons by enzyme digestion and mechanical separation is suitable for the research of patch-clamp technique.This method is simple and cost-saving.
