低氧对人脐带间充质干细胞成脂肪分化的影响

摘    要:
目的:研究低氧(2%O2)对人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUCMSCs)成脂肪分化的影响,为脂肪组织工程种子细胞的筛选提供实验依据。方法:培养条件有低氧(2%O2)、常氧(20%O2)、10%FBS完全培养基(GM)、10%FBS成脂培养基(DM);将第3代hUCMSCs根据培养条件分为3组,对照组(常氧+GM5天,常氧+DM2周)、实验组1(低氧+GM5天,低氧+DM2周)、实验组2(低氧+GM5天,常氧+DM2周);在诱导后2周分别进行油红O染色并观察其成脂效率。结果:与对照组相比,实验组1脂肪样细胞数量明显减少,实验组2脂肪样细胞数量明显增加。结论:将hUCMSCs预先进行低氧处理再转换为常氧培养可显著提高其成脂分化能力。

Abstract:
Objective To investigate the effect of hypoxia on the adipogenic defferentiation potential of human umbilical cord mesenchymal stem cells(hUCMSCs),and provide experimental evidence for selecting seeded cells of adipose tissue engineering.Methods The cultural conditions includes hypoxia(2%O2),normoxia(20%O2),10% FBS growth medium(GM),10% FBS adipogenic defferentiation medium(DM);hUCMSCs were cultured in vitro and divided into three groups according to the cultural conditions: control group(normoxia + GM 5d,normoxia + DM 2 weeks),experimental group 1(hypoxia + GM 5d,hypoxia + DM 2 weeks),experimental group 2(hypoxia + GM 5d,normoxia + DM 2 weeks).After 2 weeks of culture,the three groups were stained by oil red O.Results Compared with control group,the number of the fat-like cells decreased obvously in experimental group 1,but the number of the fat-like cells increased significantly in experimental group 2.Conclusions hUCMSCs pre-exposed to hypoxia when switched to normoxia exhibited significantly higher adipogenic defferentiation capacity.
