头针对急性脑缺血再灌注大鼠NGF表达的实验研究

目的观察头针对局灶性脑缺血再灌注模型大鼠神经生长因子表达的影响,探讨头针治疗脑缺血的可能机制。方法将70只健康Wistar雌性大鼠随机分为假手术组、模型组、头针组(模型组和头针组再根据缺血再灌注时间的不同,随机分为7d、14d、28d共6个亚组)。采用大脑中动脉线栓法制备大脑中动脉闭塞(MCAO)再灌注模型,选取顶颞后斜线,顶颞前斜线,进针后接韩氏穴位神经刺激仪,疏密波,频率2Hz/100Hz,强度2mA,每次20min,每天1次。应用神经功能缺损评分(neurological severity score,NSS)、免疫组织化学法观察脑缺血再灌注各时间点头针对模型大鼠神经功能缺损、缺血区海马齿状回NGF含量的影响。结果头针组与模型组各时相上的组间比较:(1)NSS指标:第7天、14天、28天头针组的NSS显著低于模型组(P0.05,P0.01);(2)缺血区海马齿状回NGF表达情况:各时间点头针组和模型组与假手术组比较有显著性差异(P0.01),缺血再灌注7d、14d、28d,头针组各时间点均较模型组显著升高(P0.01)。结论头针可通过增加NGF的含量来促进急性脑缺血再灌注损伤过程中神经干细胞的修复,实现对缺血脑组织保护的目的 。
