摘 要: | 目的:探讨环孢菌素A(CsA)、汉防己甲素(Tet)及两者联合应用对人白血病耐药细胞系K562/A02多药耐药的逆转作用。方法:采用M1Tr法测定柔红霉素(DNR)对细胞的半数抑制量(IC50),流式细胞仪检测细胞凋亡率和细胞内DNR浓度。结果:DNR对K562细胞和K562/A02细胞IC50分别为0.27和21.22mg·L^-1;CsA(1mg·L^-1)及Tet(0.1mg·L^-1)单独和联合处理K562/A02后,DNR的IC50分别为7.83、5.32和2.32mg·L^-1;CsA和Tet对DNR作用K562的IC50无明显影响;K562/A02细胞48h凋亡率为2.19%,DNR(1mg·L^-1)作用K562/A02细胞凋亡率为2.70%,CsA和Tet单独及联合处理后凋亡率分别为10.27%、17.64%和52.79%;两药处理后细胞内化疗药物DNR浓度亦明显增加。结论:CsA及Tet均可逆转耐药,且联合作用逆转效果更强。

Abstract: | To study the reversible effect of cyclosporine A( CsA), tetrandrine(Tet) and their combination on multidrug resistance of cell line K562/A02. Methods The IC50 of DNR was determined by MTT assay. The apoptotic changes and the intracellular DNR concentration were observed by flow cytometry. Results Treating K562/A02 cells with CsA( 1 mg·L^-1 ) ,Tet(0.1 mg·L^-1) and both for 48 hours partially restored the sensitivity of K562/A02 cells to DNR, but had no effect on K562 cells;K562/A02 cells showed apoptotic characteristics after treated with CsA and both for 48 hours; CsA and Tet (alone or combination) elevated the intracellular DNR concentration in K562/A02 cells. Conclusion Multidrug resistance can be partially reversed by CsA or Tet, the combination of both drugs shows a great reversal effect. |
