大鼠脊髓横断损伤后不同时间神经营养素表达的变化

摘    要:目的:探讨大鼠脊髓全横断后不同时间神经营养因子表达的变化。方法:30只成年健康SD大鼠,随机分为正常组(手术对照组)和脊髓全横断1d、3d、7d、14d、21d组,共6组,每组5只。采用免疫组织化学ABC法结合图像灰度分析法,检测脊髓全横断后各组神经生长因子(NGF)、脑源性神经营养因子(BDNF)和神经营养素-3(NT-3)表达的变化。结果:脊髓全横断后手术对照组NGF、BDNF、NT-3的表达较少,损伤后表达明显增加,NGF、BDNF和NT-3在损伤后1-7d表达的阳性细胞数逐渐增加,与对照组比较差异有显著性(P<0.01),7d达到最高;NGF和NT-3的高表达一直维持到21d,而BDNF在损伤后14d恢复到正常。结论:在脊髓横断损伤中,神经营养因子NGF、BDNF和NT3的表达增加,可能起着保护作用。

Abstract:Objective To explore the expression of neurotrophins in different interval after transected spinal cord of rat.Methods 30 Sprague Dawleys rats were randomly divided into 6 groups: normal control group,after transsected spinal cord operation for 1day,3days,7days,14days and 21days groups.The transected spinal cord model was made by the operation.The immunohistochemical method was used to detect the expression changes of NGF,BDNF and NT-3 in each group,and Image Analysis System was used to the quantitative analysis of experssion of NGF, BDNF and NT-3.Results After trasected spinal cord in rat,the expression of neurotrophins had some signifcant changes in the different time interval.Compared with the control group,the postive cells of NGF,BDNF and NT3 were significantly increse form 1d to 7d(P<0.05),and reach the peak in the 7d.NGF,and NT3 were significantly different in 21d after transected spinal cord compared with control group,and the expression of BDNF was no significant difference between 14d group and control group.Conclusion The expression of neurotrophins NGF,BDNF and NT-3 increase in transected spinal crod,they may have some protective effects on spinal cord injury.
