筋脉通对糖尿病大鼠坐骨神经NADPH 氧化酶p22-phox亚基表达的影响

摘    要:目的观察中药筋脉通对糖尿病大鼠坐骨神经NADPH氧化酶p22-phox亚基表达的影响。方法将STZ诱导的糖尿病大鼠随机分为模型组,维生素C组,筋脉通小、中、大剂量组,并设立正常对照组,连续灌胃16周。检测各组治疗前及治疗后4、8、12、16周的体重、血糖;测定第16周时大鼠机械痛阈值;采用免疫组化法测定大鼠坐骨神经NADPH氧化酶p22-phox亚基的表达,并进行半定量分析。结果与正常组相比,各组糖尿病大鼠体重均显著下降(P〈0.01),血糖均明显升高(P〈0.01);与模型组比较,各治疗组大鼠体重及血糖在各时间点均无显著差异(P〉0.05)。与正常组相比,模型组、Vc组、筋脉通小、大剂量组机械痛阈值显著降低(P〈0.01);各治疗组机械痛阈值较模型组均明最升高(P〈0.01);与VC组比较,筋脉通中剂量组机械痛阈值显著升高(P〈0.01)。与正常组相比,模型组及各治疗组p22亚基的IOD值明显升高(P〈0.05,P〈0.01);各治疗组p22哑基的IOD值较模型组均显著降低(P〈0.01);与VC组比较,筋脉通中、大剂量组p22亚基IOD值显著降低(P〈0.01,P〈0.05)。结论筋脉通能显著降低大鼠坐骨神经NADPH氧化酶p22-phox亚基的表达。

Effect of Jin Mai Tong on p22 - phox Subunit Expression of NADPH Oxidase in Sciatic Nerve of Diabetic Rats

Abstract:To observe the effect of JinMaiTong(JMT) on p22 - phox subunit expression of NADPH Oxidase in sciatic nerve of DM rats. Methods Diabetic rats induced by streptozotocin(STZ) were randomly divided into model group, vitamin C group, low- dose, moderate -close and high -dose JMT group in addition to control group. All rats were given intragastrie administration for 16 weeks. Body weight and blood glucose were detected before the treatment and at the 4th, 8th, 12th, 16th week after treatment. Pain threshold was measured at the 16th week. The expression of p22 - phox subunit in sciatie nerves was quantified by SABC immunohisto- chemical method and then analyzed with semi - quantitative method. Results Compared with control group, the body weight level of STZ -DM rats decreased significantly (P 〈0.01 ) while the scale of blood glucose raised strikingly (P 〈0.01 ). In all treatment groups,there were no significant differences in body weight levels or blood glucose levels compared with model group at any time ( P 〉 0.05 ). Compared with control group, the pain threshold level of model group, VC group,low - dose and high - dose JMT group reduced distinctly (P 〈 0. O1 ), while the integrated option density(IOD) scale of p22 -phox subunit expression of model group and treated groups increased obviously( P 〈 0.05 ,P 〈 0.01 ). The threshold scale of all treated groups was higher than model group( P 〈 0.01 ), while the IOD level was lower than model group(P 〈 0.01 ). Compared with VC group, moderate - dose JMT group experienced a raise of the threshold level (P 〈 0.01 ) , while moderate- dose and high -dose JMT group reduced in the IOD scale of p22 -phox subunit expression( P 〈 0.01 ,P 〈 0.05). Conclusion JMT is effective in reducing p22 - phox subunit expression level of NADPH oxidase in sciatic nerve of DM rats.
