摘    要:目的疱疹病毒相关性泛素特异性蛋白酶(herpes associated ubiquitin specific proteas,HAUSP)是泛素特异性修饰酶家族成员之一,能催化水解泛素分子c.末端的肽链连接,起到去泛素化作用。已有研究表明其参与调节多种癌/抑癌蛋白的调控,影响转录活性,与肿瘤的发生发展关系密切,但在胶质瘤组织中的研究国内外均尚未见报道。检测HAUSP在不同级别人脑胶质瘤中的表达及其与肿瘤恶性程度之间的关系,探讨其在胶质瘤发病过程中的作用。方法采用免疫组织化学染色法,免疫印迹法(WB),实时荧光定量-聚合酶链反应(RT—PCR)方法,从蛋白和mRNA水平检测80例不同病理级别胶质瘤组织和10例正常脑组织标本中HAUSP的表达。结果HAUSP在正常脑组织中低表达,而在胶质瘤组织中的表达随着病理级别的升高而增多,且具有统计学意义。结论HAUSP与胶质瘤级别呈正相关性,HAUSP可作为胶质瘤诊断和治疗的潜在靶点。

Abstract:Objective HAUSP is one of the members in the ubiquitin specific processing family of deubiquitinating enzymes (DUBs). The deubiquifinylatian may play the critical roles to regulate many proteins involved in the cell cycle, as well as tumor suppressor and oncogenes. However, the expression pattern of HAUSP in gliorm patients is still unclear. The purpose of the study is to investigate expression and significance of HAUSP in gliotms. Methods A total of 80 gliorm specimens and 10 normal control samples were collected. Immunochemistry assay, quantitative real-time Polymerase Chain Reaction (RT-PCR) and Western blot analysis were carried out to explore the expression of HAUSP. And the relation between HAUSP expressions and the clinicopathological parameters of glioma patients was analyzed. Results hmnanohistoehemistry showed that HASUP expression was increased in glioma. HAUSP mRNA and protein levels were both significantly higher in glionma samples compared to control (P 〈0. 05). In addition, real-time PCR, inmmnohistoehemistry and Western blot showed HAUSP expression was increased with the increase of ghoma mahgancy. Conclusion The expression of HAUSP is positively correlated with the glioma malignancy. HAUSP may play an important oneogenie role in glioma progression, and it is a potential diagnostic and therapeutic target.
