自然杀伤细胞在四氯化碳诱导的急性肝衰竭中的作用

摘    要:目的探讨自然杀伤细胞(NK)在急性肝衰竭进展过程中的作用。方法 40只Balb/c小鼠随机分为实验组和对照组,实验组腹腔注射四氯化碳诱导急性肝衰竭,对照组则注射橄榄油。分离小鼠外周血、肝脏、脾脏的淋巴细胞,用流式细胞仪检测其NK细胞的比例及相关受体表达情况。结果急性肝衰竭小鼠肝脏NK细胞比例较对照组明显升高,差异有统计学意义[(37.2±2.8)%vs(15.3±1.5)%,P=0.0004],外周血以及脾脏中NK细胞比例均较对照组下降。急性肝衰竭小鼠肝脏活化性受体NKP46较抑制性受体NKG2A表达升高更明显,差异有统计学意义[(18.7±1.5)%vs(4.9±2.3)%;t=3.95,P=0.016)]。结论肝脏局部NK细胞在药物诱导的急性肝衰竭中发挥重要的作用。

Abstract:Objective To investigate the effect of nature killer cells (NK) in mice with acute liver failure (ALF). Methods A mouse model of acute liver failure was established by intraperitoneal injection of a sublethal dose carbon tetrachloride. Lymphocytes were separated by Lymphocyte separation medium from liver, peripheral blood and spleen. The proportion and surface receptor expression of NK cells in liver, spleen and blood were analyzed by flow cytometry. Results The NK cells of liver in ALF mice were significantly higher than that in control group [ (37.2±2.8)% vs (15.3±1.5)%, P=0.0004) ]. The NK cells of peripheral blood and spleen in ALF mice were lower than those in control group. Expression of activatory receptor NKP46 in ALF liver NK cells increased more obviously than inhibitory receptor LY49A[ (18.7±1.5)% vs (4.9±2.3)%;t=-3.95,P=0.016]. Conclusion Liver NK cells play an important role in CCL4 induced acute liver failure.
