羊膜上皮细胞移植对脑缺血缺氧新生大鼠脑神经元及生长相关蛋白43的影响

摘    要:目的探讨羊膜上皮细胞移植对缺氧缺血新生大鼠脑生长相关蛋白43和神经元烯醇化酶(N SE)表达情况的影响。方法 7 d龄大鼠随机分为假手术组、模型组和移植组,移植组和模型组经侧脑室注入1×106BrdU标记人羊膜上皮细胞悬液和PBS治疗;假手术不治疗,移植后1、2和3周观察大鼠神经功能的变化,免疫组织化学法检测脑BrdU阳性细胞、生长相关蛋白43、N SE的表达。结果移植组和模型组大鼠的神经功能评分明显改善,移植后第3周脑切片检出BrdU阳性细胞存在,模型组生长相关蛋白43和N SE在第3周的表达均低于假手术组,移植组较模型组增加。结论羊膜上皮细胞移植入脑缺血缺氧大鼠脑内后,可减轻缺氧缺血大鼠神经元的损伤,G AP-43和N SE蛋白表达增强,促进神经轴突的生长与延伸,从而改善神经功能。

Effect of transplantation human amniotic epithelial cells on the neuron and growth associated protein 43 of neonatal rats after hypoxic-ischemic encephalopathy

Abstract:【Objective】To study the expression of growth associated protein 43(GAP-43) and neurone specific enolase(NSE),after transplantation of human amniotic epithelial cells(hAECs) into the brain of neonatal rats with hypoxic-ischemic injured.【Methods】7 days old rats were randomly divided into sham operation control,model group,transplantation group.The 1×106 hAECs marked with BrdU and PBS were respectively injected into cerebral ventricle of transplantation group and model group,sham operation group did not treated.Neurological were observed at 1,2 and 3 weeks after transplantation.BrdU-positive cells and the expression of GAP-43 and NSE proteins were in brain by immunohistochemical following transplantation.【Results】Compared with model group,neurological function were significantly improved in the transplantation group,BrdU positive cells were detected in transplantation group rat brains at 3 weeks following transplantation,GAP-43 and NSE proteins in model group were significantly lower than sham operation group,cell group and the supernatant group were higher than that of the model group at 3 weeks following transplantation.【Conclusion】HAECs of injected into the cerebral ventricles can could reduce the hypoxic-ischemic injury to neurons,growth associated protein 43 and NSE expression was significantly increased,which promotes axonal growth and extension and final improve neurological function.
