大黄水提物对成年大鼠子宫及卵巢的影响

摘    要:
目的: 观察长期灌胃大黄对雌性大鼠子宫及卵巢的影响。 方法: 8周龄180~220 g 雌性Wistar大鼠90只,随机分为正常对照组、大黄水提取物1.0,2.0 g·kg-1组、各剂量组再分为灌胃30,40,50,60 d组,每组10只,计算大鼠子宫、卵巢指数,观察子宫和卵巢的组织形态学变化。 结果: 大黄水提取物1.0 g·kg-1给药60 d组子宫和卵巢指数分别为(0.21±0.058),(0.07±0.001),均显著低于正常对照组(0.26±0.038),(0.07±0.007)(P<0.05);大黄水提物2.0 g·kg-1给药40,50,60 d组子宫指数分别为(0.20±0.045),(0.19±0.054),(0.19±0.041),显著低于正常对照组(0.26±0.038)(P<0.05或P<0.01),大黄水提物2.0 g·kg-1给药30,40,50,60 d组卵巢指数分别为(0.068±0.008),(0.065±0.007),(0.065±0.005),(0.061±0.006),均显著低于正常对照组(0.07±0.007)(P<0.05或P<0.01);组织形态学显示,2.0 g·kg-150,60 d组大鼠子宫内膜水肿,腺体减少;卵巢萎缩,卵泡数减少,卵泡内细胞排列不规则。 结论: 长期应用大黄可影响成年雌性大鼠的生殖功能,并有明显的剂量/时间依赖关系。

Abstract:
Objective: To observe the long-term effect of Rhubarb on the uterus and ovary in female rats. Method: Ninety 8-week-old female rats (180-220 g) were randomly divided into nine groups:normal control group, and investigation groups (n=10 each, administration of Rhubarb 1.0,2.0 g·kg-1 for 30, 40, 50, 60 d respectively).Then the index of uterus and ovary were calculated, and the morphological changes in uterus and ovary were observed. Result: The indexes of uterus and ovary in Rhubarb 1.0 g·kg-160 d group were lower than that in the control group obviously (P<0.05);the indexes of uterus in 2.0 g·kg-1 groups (40, 50, 60 d) were lower than that in the control group obviously (P<0.05 or P<0.01). While, the indexes of ovary in 2.0 g·kg-1 groups (30, 40, 50, 60 d) were lower than that in the control group obviously (P<0.05 or P<0.01). The pathological examination showed that endometrial edema and glandular reduction, ovary atrophy, follicles reduction, and follicle cells irregular arrangment in 2.0 g·kg-1 groups (50, 60 d). Conclusion: The long-term use of Rhubarb shows a certain degree of reproductive toxicity in female rats, and its impact is clear dose/time dependent.
