三苯氧胺联合顺铂抑制人卵巢癌细胞HO-8910增殖的实验研究

目的:研究三苯氧胺(TAM)和顺铂(DDP)对人卵巢癌细胞生长的影响,验证两种药物联合应用治疗人卵巢癌的有效性。方法:以体外培养的人卵巢癌HO-8910为研究对象,不同剂量的TAM和DDP联合作用于人卵巢癌细胞HO-8910,用四甲基偶氮唑蓝比色(MTT)法检测细胞生长抑制率,用免疫组化SABC法检测增殖细胞核抗原(PCNA)在细胞中的表达。结果:TAM和DDP联合用药组与单用DDP组相比,大剂量TAM5.0μmol/L合用DDP及中剂量TAM1.0μmol/L合用大剂量DDP3.3μmol/L细胞生长抑制率有显著性差异(P<0.05);TAM浓度(≥1.0μmol/L)合用DDP时PCNA阳性率有显著差异(P<0.05)。结论:TAM和DDP联合用可有效抑制HO-8910细胞生长及增殖。

Inhibition of Proliferation of Hmnan Ovarian Cancer Cell Line HO-8910 in vitro with Tamoxifen Combined with Cisplatin

Objective: To investigate the effect of tamoxifen combined with cisplatin on the growth of human ovarian cancer cell,and to insepect the efficacy of the combined treatment of human ovarian cancer with tamoxifen and cisplatin.Methods: Human ovarian cancer cell line HO-8910 was studied.Different doses of TAM and DDP was combined in human ovarian cancer cell line HO-8910.The growth inhibition ratio of the cell was detected with MTT,the expression of Prolifering cell nuclear antigen(PCNA) in HO-8910 was detected with immunohistochemical stining(SABC).Results:compared with positive control group high-dose TAM(5.0μmol/L)combined with DDPand intermedial dose TAM(1.0μmol/L) combined with high-dose DDP(3.3μmol/L)had significant difference in the growth inhibition ratio of the cell(P<0.05);TAM(≥1.0μmol/L)combined with DDP had significant difference in the expression of PCNA(P<0.05).Conclusion: TAM combined with DDP suppressed the growth and proliferation of the cell HO-8910 efficiently.
